Isolation and Identification of Pathogens of Poplar Stem Diseases

Since 2017, Populus cathayana in Shuangshu Nursery, Huzhu County, Qinhai Province has suffered from extensive necrosis of branches, yellow pith tissue, cracks bark to severe cases, brown mucus outflow summer, yellowish pith, and no obvious disease. In this study, symptom observation, specimen collection, pathogen isolation culture, morphological observation molecular biological identification were carried out on poplar branch diseases. After the was isolated, cultured purified, a pure bacterial strain H1 obtained its pathogenicity tested. The results showed that pathogenic bacterium caused disease plants, which consistent with symptoms field. Through sequence analysis, identified as Pantoea agglomerans, can cause canker.
